Electroporation
- Mix 10 ng of plasmid with 50 µl of electrocompetent E.Coli cells prepared as described in protocol 1).
- Transfer in a prechilled 2 mm electroporation cuvette
- Perform electroporation. Setted parameters: Capacitance: 25 µF; Voltage: 2.5 kV; Resistance: 200 ohms.
- Immediately after electroporation add 70 µl of sterile 2xTY- 1%Glucose and plate in a 20 cm diameter plate of with suitable antibiotics by gently spreading with a sterile glass loop
Efficiency of transformation should be > 5x108 transformants/µg
of plasmid DNA.
