Preparation of Media for Bacterial growth

There are mainly two media used in the lab: LB or 2xTY. For protein expression 2xTY is used.  The media is normally prepared as a liquid suspension and then autoclaved.
For big cultures, the media is often prepared directly in the culture flasks.

LB:
Dissolve 10 g tryptone, 5 g yeast extract and 10 g NaCl in 800 ml dH2O. Adjust the pH to 7.0 with 1M NaOH. Adjust the volume to 1 liter with dH2O. Sterilize by autoclaving.

2xTY:
Dissolve 16 g tryptone, 10 g yeast extract and 5 g NaCl in 800 ml dH2O. Adjust the pH to 7.0 with 1M NaOH. Adjust the volume to 1 liter with dH2O. Sterilize by autoclaving.

Note:
For 2xTY there is also the possibility to use an already formulated 2xTY Broth produced by Bio101. One can use the pre-formulated powder or capsules. Simply weigh 31 g powder or count 31 capsules per liter, mix with dH2O and autoclave. The powder or the capsules do not have to be dissolved before autoclavation.
